About me

My teaching is in biochemistry and my research area is the molecular epidemiology of infectious agents, particularly the use of genotyping tools for improving detection methods and for studying transmission dynamics. My group works on the molecular epidemiology of enteric parasites such as Cryptosporidium and Giardia and blood borne parasites such as Babesia, Theileria and Trypanosomes. We are also interested in novel therapeutics for pathogens.

Research areas

Our research has concentrated on the molecular detection and epidemiology of vector and water-borne pathogens (http://www.murdoch.edu.au/Research-capabilities/Vector-and-Waterborne-Pathogens-Group/). This includes some of the following areas:


  • Molecular epidemiology/taxonomy of protozoan parasites

We are currently working on an ARC-Funded project entitled: Innovative approaches to understanding and limiting the public health risks of Cryptosporidium and Giardia in animals in Australian catchments (Linkage Project LP130100035).

Cryptosporidium & Giardia (C&G) are the major public health concern of water utilities & are responsible for disease in a wide range of hosts, 95.5% of waterborne protozoan outbreaks worldwide, can be fatal in the immunocompromised and are resistant to chlorine. This proposal will target a key knowledge gap; the lack of quantitative data on human infectious sp. It will analyse C&G in animals and human sewage in catchments across 3 states (WA, NSW and Qld), identify the species/ subtypes, the percentage viability and develop improved modelling and quantitative microbial risk assessments (QMRA). The project will result in the development of more targeted, cost-effective measures to minimise exposures to infections, accurate risk assessment, & scientific catchment management.

Dr. Andrea Paparini is the post-doctoral research fellow on this project, who co-supervises the PhD student Alireza Zhaedi with Prof. Ryan and Prof. Ian Robertson.

Prof. Ryan also collaborates with A/Prof. Alan Lymbery on the molecular epidemiology of Cryptosporidium and other parasites in fish.

Other current research includes a Meat and Livestock Australia (MLA)-funded project entitled “Impact of bacteria and coccidia on scouring & productivity in sheep”. Scouring and dag in sheep and subsequent breech blowfly strike is a major and widespread economic and animal welfare problem for the Australian sheep industry, yet the causes of scouring in sheep remain poorly understood.  This project involves the development of molecular tools to examine if scouring and production loss is associated with particular protozoan or bacterial species/genotypes and with particular management systems under Australian conditions. The project is run by Dr. Rongchang Yang.

We also have a MLA-funded project in collaboration with A/Prof. David Millar working on a project entitled “Causes of scouring and ill-thrift in Rangeland goats”.

The Australian goatmeat industry has experienced strong growth over the past 20 years and this growth has been largely supported by the sale of goats derived from rangeland or extensive production systems. Of the 1.5 million goats now slaughtered annually, approximately 90 percent are rangeland goats. Relocating goats into close confinement situations can disrupt their social structure and dominance behaviour. In addition, poor nutrition, lack of shelter, overcrowding and excessive handling can lead to further stress, which aids the development of disease. Little is known about their prevalence and contribution to scouring in rangeland goats in Australia. This project is being undertaken by Khalid Al Habsi as part of his PhD thesis and will investigate parasitic and bacterial causes of scouring in rangeland goats.

Sylvia Afriyea has just commenced her PhD in Jan 2014 working on a project entitled “Enhancing livestock productivity through the molecular epidemiology of Cryptosporidium as emerging zoonoses in Ghana”.

  • Vector-Borne diseases

The molecular epidemiology of tick-transmitted blood parasites and bacterial infections of companion animals in Australasia is an area that is poorly understood and has important implications for AQIS (Australian Quarantine and Inspection Service). Dr. Ryan Jefferies who completed his PhD in this area under the supervision of A/Prof. Peter Irwin and Professor Ryan, developed a DNA-based multiplex diagnostic for tick-transmitted blood parasites and Rickettsia infections. This test has now been made available to AQIS to assist them in maintaining Australia’s largely exotic pathogen-free environment. Dr Gunn Kaewmongkol recently completed a PhD thesis on the molecular characterisation of Bartonella spp and identified several new species in Australian native and feral animals. In 2012, A/Prof. Irwin and Dr. Paparini were involved in the identification of zoonotic babesiosis (Babesia microti), known previously only in the Northern Hemisphere, in a person from the NSW south coast with no travel history.

We are currently working on an ARC-funded project entitled “Troublesome Ticks: A new molecular toolkit to investigate tick-borne pathogens in Australia”.  Recreational and employment activities of humans in rural regions, often with companion animals, provide ideal environments for transmission of vector-borne diseases from wildlife. Currently in Australia there is community concern and uncertainty about the occurrence, diversity and distribution of tick-associated pathogens. The primary objective of this project is to systematically characterize and map potentially zoonotic tick-borne organisms (bacteria, rickettsiae and protozoa) using a new molecular toolkit. This project will create important data about vector-pathogen-host ecologies in Australia and provide new understanding about the potential risk for humans and companion animals of tick-borne disease, nationwide. Trypanosomiasis is a complex debilitating and often fatal disease caused by infection with one or more of the pathogenic arthropod transmitted protozoan parasites of the genus Trypanosoma. Trypanosomes have a global impact on the health, survival and economic development of millions of people and animals in temperate, tropical and sub-tropical regions. Little is known of the prevalence and life-cycle of trypanosomes in mammals native to Australia. A current PhD project being undertaken by Jill Austen under the supervision of Prof. Una Ryan and Dr Tony Friend (Department of Environment and Conservation) examines novel Trypanosoma sp. identified in blood from the critically endangered Gilbert’s potoroo (Potorous gilbertii) and the quokka (Setonix brachyurus) in Western Australia. Jill recently identified the tick vector for the trypanosome species infecting the Gilbert’s potoroo. Another recently gradated PhD student, Linda McInnes identified several new species of Trypanosomes in Koalas under the supervision of Prof. Una Ryan. A new PhD student, Amanda Durante, supervised by Prof. Una Ryan, A/Prof. Irwin and A/Prof. Kris Warren, is looking at enteric and blood-borne parasites in marsupials.

  • Other projects

Dr. Andrea Paparni is working on an ARC Linkage grant in collaboration with Dr. Paul Monis (AWQC) entitled “Innovative approaches to managing and understanding taste and odour in drinking water systems”. Cyanobacterial blooms are a costly and undesirable result of land use, agriculture and climate change affecting the quality of global source-water. Cyanobacteria can produce deadly toxins and also metabolites like geosmin, tainting water with unpleasant taste and odour (T/O). This project exploits the latest groundbreaking discoveries, to characterize the major physiological, biochemical, and molecular traits of geosmin producers, the factors commonly associated with production, and the molecular regulation of biosynthesis. The outcomes will improve effective and sustainable management of water resources, provide novel biomolecular assays for early warning, and an improved understanding of the ecological controls of T/O production.

In collaboration with colleagues at Curtin University, Prof. Ryan and Dr. Paparini is part of an ARC-Linkage project entitled “Understanding wastewater treatment technologies for alternative water use: transformation of inorganic and organic nitrogen” – LP130100602. Reuse of treated wastewater for non-potable purposes is an important water management strategy in many parts of rural Australia, however industry uptake is hampered by a lack of knowledge of the chemical and microbial risks of alternative uses of treated wastewater. This study will provide a new understanding of the chemistry of inorganic and organic nitrogen removal in rural wastewater treatment systems, and use novel molecular tools to profile the microbial community in wastewater before and after treatment. Improved knowledge of wastewater treatment process efficacy will improve compliance with health guidelines, protect public and environmental health, and enable an increased uptake of alternative water use options in rural Australia.

Current projects

  • Research on tick-transmitted Piroplasmida and trypanosomes is ongoing and has included the identification of a novel Theileria sp. in the critically endangered Gilbert’s potoroo (Potorous gilbertii) and declining species of threatened Australian marsupial, the brush-tailed bettong (Bettongia penicillata). This research is currently being conducted by Dr. Andrea Paparini.

 

  • Molecular characterization of the enteric protozoan parasites, Cryptosporidium and Giardia in wildlife. This research is currently being conducted by Dr. Andrea Paparini and Dr. Rongchang Yang.
